The Role of ERK1/2 in Astragalus Polysaccharide-induced Apoptosis in HepG2 Cell Line
Objective: Although astragalus polysaccharide(AP) is known to have apoptotic effect in many cancer cells
the mechanisms are still poorly understood. We aim to test whether apoptotic effect of astragalus polysaccharide is regulated by the ERK1/2 pathway in HepG2 cells. Method: Cells were divided into 4 groups and treated with different pharmaceuticals:control

cell viability measurement
TUNEL assay and mitochondrial potential analysis
caspase-3 activity measurement. Result: Astragalus polysaccharide has apoptotic responses in HepG2 cells
including mitochondrial depolarization and the apoptosis-specific proteins up-regulation
Bcl-2 down-regulation. Conclusion: These results suggest that ERK1/2 pathway is necessary for the effects of astragalus polysaccharide in inhibiting cell proliferation of HepG2 cells.
