Differerntiation of Mesenchymal Stem Cells into Myofibroblasts and the Impact of Yiguanjian
Objective: To observe the differentiation of bone marrow mesenchymal stem cells into myofibroblast in vitro and the impact of Yiguanjian. Method: Bone marrow mesenchymal stem cells were separated
cultured and identified
transforming growth factor β1 (TGF-β1 5 μg·L-1) was added to the third-generation cells to induce differentiation. Bone marrow mesenchymal stem cells were divided into normal group
TGF-β1 induced group
TGF-β1 induced plus Yiguanjian group. 2 weeks later
bone marrow mesenchymal stem cells were harvested and detected using immunofluorescence and real-time quantitative PCR. Result: The result of bone marrow mesenchymal stem cells’ identification showed the strong expression of CD44 and the weak expression of CD29. After TGF-β1 induced
the results of immunofluorescence and real-time quantitative PCR showed that the group of TGF-β1 induced the expression of smooth muscle ectin(α-SMA)
the expression of α-SMA in the group of TGF-β1 induced plus Yiguanjian was significantly reduced
the difference was statistically significant (P <0.05). Conclusion: Bone marrow mesenchymal stem cells can differentiate into myofibroblasts using TGF-β1 induction
Yiguanjian can prevent the bone marrow mesenchymal stem cells’ differerntiation to myofibroblast.
