Determination of Rutin from Leaves of Gathered in Different Areas and Different Periods by HPLC-UV
Objective: To study the content of rutin from the leaves of Ziziphus jujuba gathered in different areas and different periods for ascertaining the best harvest time and providing rational basis for the resource development. Method: The assay was performed on a Diamonsil C18 (4.6 mm×200 mm
5 μm) column by an isocratic elution with methanol-0.1% acetic acid(34 ∶66) at a flow rate of 1.0 mL·min-1. The detection wavelength was at 257 nm.The column temperature was kept at 30 ℃. Result: Rutin was linear in the range of 0.221 6-2.216 μg (r=0.999 9). The average recovery ( n = 6) was 99.40% (RSD 2.42%). The content of rutin reached its maximum yields in May and June
however
the content drop down in July and August and rise again in September and October. Conclusion: This method was accurate
simple and stable. The content of rutin changing with time showed a U trend. It provided reference for quality evaluation criteria of the leaves of Z. jujuba
