Comparison of Protein Extraction Methods of Trichosanthis Fructus
Objective: To determine the protein extraction method of Fructus Trichosanthis. Method :The protein yield
protein purity and polyacrylamide gel electrophoresis(PAGE) patterns were determined and analyzed. Result: More proteins were extracted by the method of protein extracted liquid and trichloroacetic acid(TCA)-acetone
but their bands were not clear and the resolution was low because of more impurities extracted.It waseasy to produce high-abundance proteins and interfered in the electrophoresis resolution.Protein yield and band numbers by Tris-base/acetone were the lowest.Besides
the protein content by commonly used direct extraction was low
but with less impurity
clearer bands and highest resolution. Conclusion: Direct extraction is the most suitable method for PAGE and more protein can be extracted by protein extracted liquid extraction method.
