载气流速1.6 L ·min-1。进样量10 μL。 结果 :黄芪甲苷在0.498~9.96 μg呈良好的线性关系
r=0.9998;平均回收率为98.05%

在常温下8 h内稳定性较好。 结论 :HPLC-ELSD法简便快速
准确
重复性好
结果可靠
可作为芪苓益肝颗粒的质量控制方法。
Abstract
Objective: To establish a determination method of the content of astragaloside in Qiling Yigan Granular formulation by HPLC-ELSD. Method: The content of astragaloside in Qiling Yigan Granular formulation was determined by HPLC-ELSD and Agilent ZORBAX SB-C18 (4.6 mm×250 mm
5 μm) column was used. The mobile phase was a mixture of acetonitrile-water (35 ∶65)
the flow rate was 1.0 mL ·min-1
column temperature was of 25 ℃. The temperature of drift tube for ELSD was 110 ℃ and the flow rate of air was 1.6 L · min-1. Result: The calibration curve of astragaloside was linear over the range of 0.498-9.96 μg (r=0.999 8)
the average recovery rate was 98.05%
RSD 0.89%. Conclusion: The method is convenient
sensitvive and accurate. It can be a method for quality control in production of Qiling Yigan Granular formulation.
