摘要

目的 :建立用固相萃取-高效液相色谱法定量大鼠灌服柴胡水提液后血浆中柴胡皂苷d的方法。方法 :大鼠灌服柴胡水提液后在不同时间点眼眶静脉取血

血浆经Waters Oasis HLB固相萃取小柱纯化

洗脱液用氮气吹扫至干

流动相溶解后进样;用YMC-Pack ODS C18色谱柱以及乙腈-水为流动相的色谱体系检测其中柴胡皂苷d的含量。结果 :柴胡皂苷d在1.28~81.60 μg ·mL-1的浓度范围内线性关系良好(相关系数r = 0.997 8)

定量下限为1.28 μg ·mL-1。相对回收率为94.6%~102.8%

日间、日内RSD均小于8%。结论 :该方法灵敏、准确

可用于大鼠血浆中柴胡皂苷d的浓度测定。

Abstract

Objective: To develop a SPE-HPLC method for the determination of saikosaponin D in rat plasma after being administered Radix Bupleuri water extract intragastrically. Method: The blood was taken from rat orbital venous at different time points

and the plasma was purified by using the Waters Oasis solid-phase extraction column. The eluate was dried with nitrogen

and then dissolved with mobile phase. The saikosaponin D was analyzed by chromatographic system consisting of YMC-Pack ODS C18column and acetonitrile-water as mobile phase. Result : Saikosaponin D was linear in range of 1.28-81.60 μg ·mL-1 (r = 0.997 8). And the quantitative limit was 1.28 μg ·mL-1. Relative recovery was 94.6%-102.8%; And the within-day precision and between-day precision were less than 8% (RSD). Conclusion: This method is sensitive

accurate

and can be used for determination of saikosaponin D in rat plasma.
