Analysis of Monosaccharide Composition in Astragalus Neutral Polysaccharides by High Performance liquid Chromatography
Objective:To analyst monosaccharide composition in astagalus neutral polysaccharides(ANP) by high performace liquid chromatography(HPLC).Method: Orthogonal design was used to optimize the hydrolytic condition of ANP by trifluoroacetic acid(TFA)
the monosacchride composition in hydrolysate was assayed directly by HPLC-RID method or HPLC-DAD method after precolumn-derivation with 1-phenyl-3-methyl-5-pyrazolone(PMP) with six monosaccharides as control.Result: The optimum hydrolization procedure of ANP was configered with 1M TFA hydrolyzing 5 hours at 100 ℃.The results of two HPLC methods showed that the ANP consisted of glucose.Conclusion: The optimum hydrolization procedure and HPLC methods are simple and convenient and could be applied to analysis of the monosaccharide composition in ANP.
