摘要

目的:观察枳椇子乙酸乙酯提取物胶囊对大鼠肝微粒体细胞色素P450及其同工酶的影响。方法:大鼠分别灌服给予枳椇子乙酸乙酯提取物胶囊相当于生药量0.14 g/kg

0.17 g/kg和0.2 g/kg

连续10 d

用紫外分光光度法测定细胞色素P450及其同工酶的活性。结果:枳椇子乙酸乙酯提取物胶囊对细胞色素P450及NADPH-细胞色素C(P-450)还原酶含量无显著影响;可抑制苯胺羟化酶活性

抑制率可达31.5%;可增加氨基比林N-脱甲基酶活性

可达42.4%;而对红霉素N-脱甲基酶的活性没有明显影响。结论:枳椇子乙酸乙酯提取物胶囊对大鼠肝细胞色素P450、NADPH-细胞色素C(P-450)还原酶及红霉素N-脱甲基酶无明显作用

但对氨基比林N-脱甲基酶活性及苯胺羟化酶活性有显著影响。

Abstract

Objective:To investigate the effects of the ethyl acetate extract of Semen Hoveniae capsule on liver microsomal cytochrome P450 system in rats.Methods:The rats were given orally the extract in the doses of 0.14 g/kg、0.17 g/kg and 0.2 g/kg(equivalent to the crude herb) for 10 days respectively.The activities of cytochrome P450 enzymes were measured by UV chromatography.Results:The cytochrome P450 content

NADPH-Cyt C reductase activities and erythromycin N-demethylase activities were not affected;Aniline hydroxylase activities in liver were decreased by up to 35.1%;aminopyrine N-demethylase activities in liver were increased by up to 42.4%.Conclusions:The ethyl acetate extract of Semen Hoveniae capsule had no inhibition effect on the liver microsomal cytochrome P450

NADPH-Cyt C reductase and levels erythromycin N-demethylase in rats.However

analine hydroxylase activities and aminopyrine N-demethylase activities were significantly affected.
