Analysis of the Response Factor of Different Amino Acids Detected by HPLC-ELSD Method
Objectives:To develop the HPLC-ELSD method to detect the different amino acids and analyze the response factors of different constituents detected by ELSD.Methods:The stationary phase was Kromasil C18 column(4.6×250mm
5μm)at 30℃.The mobile phase was water contained 0.1%TFA with flow rate was 0.3mL·min.The chromatogram was recorded by ELSD with drift tube temperature was 105℃ and gas flow rate was 2.60L·min-1.The response factors were analyzed by compare the standard curves of different amino acids.Result:The difference between the slops of standard curves was found.Conclusion:The response factor of different amino acids detected by ELSD was greatly different.
