Optimization of Cellulase Hydrolysis Extraction Process of Folium Ginseng by Central Composite Design/Response Surface Methodology
Objective:To obtain the optimal cellulase hydrolysis extraction process of Folium ginseng by Central Composite Design/Response Surface Methodology.Methods: To extract ginsenosides from folium ginseng by adding cellulase digestion process.The parameters of enzyme hydrolysis action is divided into independent variables including pH value
water volume
the ratio of enzyme/the amount of herbal
and dependent variables-the extraction rates of ginsenosides
Linear or nonlinear mathematic models were used to estimate the relationship between independent and dependent variables.The response surface methodology was used to optimize the conditions of extraction.Prediction was carried out through comparing the observed and predicted values.Results:It was showed that the coefficient of correlation of second-order quadratic model was high.The correlation coefficient reached 0.940.The optimum conditions of extraction were obtained.The optimum condition was pH 4.8
24 times amount of water more than material medica
and enzyme/substrate was 40U/g.The observed values and the predicated one of extraction ratio was 6.29%and 6.37%.Conclusion:The observed values and predicted one are close to each other
which prove that the optimization of cellulase digestion extraction process of ginsenosides from folium ginseng by Central Composite Design/Response Surface Methodology is reasonable and successful.
