Effect of Buyang Huanwu Decoction and Its Active Fractions Such as Alkaloid and Glycoside on TXA and PGI of Arterial Thrombosis Model in Rats
Objective:To explore effect of Buyang Huanwu Decoction(BHD) and its active fractions such as alkaloid and glycoside extracted from BHD on thromboxane(TXA2) and prostacyclin(PGI2) of arterial thrombosis model in rats.Methods:Carotid arterial thrombosis model in rats was induced by topical ferric chloride.The model rats were administered with BHD
alkaloid
glycoside
low molecular weight heparin(LMWH) and ticlopidine respectively.Weight of thrombus and the levels of TXB2 and 6-keto-PGF1α in plasma in each group were compared.Results:Occlusive thrombus was induced
the levels of TXB2 were increased markedly(P<0.05)
the levels of 6-ketoPGF1α were lower remarkably in the model group(P<0.05).Weight of thrombus in BHD
alkaloid
LMWH and ticlopidine group was significantly lower than that of the model group(P<0.05).BHD could increase the levels of 6-ketoPGF1α(P<0.05)
but it did not influence the levels of TXB2.Alkaloid could decrease the levels of TXB_2 and increase the levels of 6-keto-PGF1α significantly(P<0.05).Glycocide
LMWH and ticlopidine had no influence on the levels of TXB2 and 6-keto-PGF1α.Conclusions:Antithrombotic actions of BHD
alkaloid
glycoside
LMWH and ticlopidine were observed.Antithrombotic actions of BHD may be related to accelerating synthesis of PGI2.Antithrombotic actions of alkaloid may be associated to inhibiting production of TXA2 and increasing synthesis of PGI2.Antithrombotic actions of glycoside
LMWH and ticlopidine was not based on its modulation on TXA2 and PGI2
