摘要

目的：建立东风桔总生物碱的含量测定方法并优选其提取工艺。方法：采用酸性染料比色法测定东风桔总生物碱含量，选择Buxifoliadine-B为指标成分，检测波长410 nm。以总生物碱提取量为指标，通过正交试验考察乙醇体积分数、乙醇用量、回流时间及提取次数对提取工艺的影响。结果：Buxifoliadine-B含量测定的线性范围0.034~0.34 g·L-1，加样回收率（99.16±1.81）%。最佳提取工艺为加10倍量80%乙醇回流提取2次，每次1.5 h；总生物碱提取量4.88 mg·g-1。结论：建立的东风桔总生物碱含量测定方法稳定可靠，优选的提取工艺方法合理可行。

Abstract

Objective: To establish a determination for total alkaloids in Atalantia buxifolia and optimize its extraction process. Method: The content of total alkaloids was determined by acid dye-colorimetry

extracting time and times on extraction amount of total alkaloids were investigated by orthogonal test. Result: The content of Buxifoliadine-B had a good linear relationship within the range of 0.034-0.34 g·L-1

extraction amount of total alkaloids was 4.88 mg·g-1. Conclusion: This determination for total alkaloids in A. buxifolia and optimized extraction process were reasonable and practicable.
