并探讨其作用分子机制。 方法: 通过MTT法测定雷公藤内酯醇对胶质瘤细胞株U87细胞增殖抑制作用

Objective: To study the anti-invation effct of triptolide for U87 glioma cells and its molecular mechanism. Method: MTT assay was used to determine the growth inhibition by triptolide in glioma U87 cells. Tumor invasion and migration were examined by Transwell chamber assay and wound-healing method. Real-time PCR was conducted to detect alteration of matrix metallopeptidase 9 (MMP-9) and Cathepsins B (CB). The alteration of nuclear factor kappa B (NF-κB) p65 expression were studied by Western blot. Result: Triptolide could significantly inhibite glioma U87 cell proliferation
reduce the cell ability of migration and invasion
down-regulating its MMP-9
CB and NF-κB p65 expression level. Conclusion: Triptolide could depress the invasion of glioma cells owing to down-regulating the level of NF-κB pathway which could inhibit MMP-9 activities
reduce the ability of hydrolyzing extracellular matrix in vitro.
