Determination of Nucleosides Compounds in Walnut Kernel from 10 Different Regions
guanosine and adenosine in walnut kernel by HPLC. Method: The HPLC method was used to determine respectively the content of cytidine
uridine
guanosine and adenosine in walnut kernel from 10 different regions. The analysis was carried out on Agilent TC-C18 column (4.6 mm×250 mm
5 μm). Methanol-10 mmoL·L-1ammonium acetate was used as mobile phase with gradient elution; the flow rate was 0.5 mL·min-1
detection wavelength was set at 260 nm and the column temperature was kept at 30 ℃. Result: The injection amounts of four nucleosides compounds over respective ranges displayed good linear relationship with the peak area and the correlation coefficients equaled or exceeded 0.999 3.The average spike recoveries for four nucleosides compounds ranged from 97% to 102%
with a RSD of less than 3%. Conclusion: The method is simple
quick
accurate and reproducible
and can be provide reference for the quality control of the walnut kernel.
