Objective: To observe the effects of different concentrations of tetrahydroxy stilbene glycoside (TSG) on the growth and proliferation of neural stem cells(NSCs) which were injured by Aβ25-35
to discuss the protective effects of TSG on the NSCs injury induced by Aβ25-35. Method: NSCs were extracted on both sides of brain cortex from the embryos of 14 days of Kunming mice pregnancy
NSCs were cultured by Aβ25-35(25 μmol·L-1) to establish cell model of Alzheimer's disease (AD) and then were cultured with TSG. All cells were divided into 5 groups
control group; model group(Aβ25-35 25 μmol·L-1)
(TSG 1×10-8mol·L-1+Aβ25-35)group
(TSG 1×10-7 mol·L-1+Aβ25-35)group and (TSG 1×10-6mol·L-1+Aβ25-35)group.Using the chemical fluorescence staining method (EDU) to detect the proliferation of NSCs; applicating WST-1 cell proliferation and cell toxicity test to detect the activity of NSCs of different time and then after cultured 7 days in vitro
