Determination of Three Active Components in Niuhuang Xiaoyanling Capsule by HPLC
Objective: To develop a reversed-phase HPLC method for the determination of three active components including geniposide
berberine hydrochloride and baicalin in Niuhuang Xiaoyanling capsule. Method: The determination was performed on a ZORBAX Eclipse XDB-C18 column (4.6 mm×150 mm
5 μm) with acetonitrile-0.2% phosphoric acid as the mobile phase by gradient elution at the flow rate of 0.8-1.0 mL·min-1. The detection wavelength was set at 240 nm for geniposide and hydrochloride baicalin
278 nm for berberine respectively. The column temperature was maintained at 30℃. Result: The calibration curves of geniposide
berberine hydrochloride and baicalin were linear in the ranges of 0.020 4-0.408 μg (r=0.999 7)
0.041 2-0.412 μg (r=0.999 8)
0.060 8-0.608 μg (r=0.999 9) respectively. The average recoveries were 99.12%
99.92% and 99.32% respectively; RSDs were 1.77%
1.24%
1.63% respectively. Conclusion: This proposed method is simple
accurate and reproducible
and suitable for the determination of the content of three analytes in Niuhuang Xiaoyanling capsule; it can be also used for quality control and evaluation of related capsule.
