Objective: To establish a HPLC method for determination of kaempferol in D. caudatum. Method: D. caudatum was extracted by reflux with the mixed solution of methanol 25% hydrochloric acid (4:1) for 1.5 hours. The chromatographic column was ThermoBDS Hyperl C18 column (4.6 mm×250 mm
5 μm)
and the mobile phase consisted of acetonitrile-0.2% phosphoric acid (30:70) with a flow rate of 1.0 mL·min-1. The detection wavelength was set at 368 nm
and the column temperature was kept at 30℃;the inject volume was 10 μL. The content of kaempferol in D. caudatum was determined by external standard method. Result: Kaempferol was linear in the concentration range of 18.94-189.4 ng (r=0.999 6)
and the average recovery of kaempferol was 105.5% (RSD 1.2%). Conclusion: The method is simple
accurate and reproducible with good stability
which can be used as quality control method for D. caudatum.
