柱温 30℃。 结果: 最大吸收波长在340 nm
HPLC在秦皮素0.08~0.48 μg(r=0.999 8)有良好的线性关系
平均加样回收率99.61%
RSD 1.41%。 结论: 建立的白蜡树子质量标准操作简便、准确快速、重复性好
并能有效控制维吾尔药白蜡树子的质量。
Abstract
Objective: To establish the quality evaluation of Fraxinus sogdiana seed. Method: A series of experiments were performed to study the impurity
including moisture
the ash
the acid-insoluble ash
heavy metal
pesticide residue and extract determination according to the Pharmacopoeia. Microscopic identification and TLC was performed to identify Fraxinus sogdiana seed. HPLC was used to determine the content of ferulic acid. Phenomenex luna C18 (4.6 mm×250 mm
4 μm) column was used. The mobile phase consisting of methanol-0.1% phosphoric acid water solution (35: 65) and UV detection wavelength at 340 nm;the flow rate was 1.0 mL·min-1;column temperature was kept at 30℃. Result: The UV biggest absorption wavelength was found at 340 nm. Fraxetin showed a good linearity in the range of 0.08-0.48 μg (r=0.999 8). The average recovery was 99.61% (n=6)
RSD was 1.41%. Conclusion: Quality evaluation methods of Fraxinus sogdiana seed were established and content limit of fraxetin was confirmed. And the methods preformed in this study were simple
accurate and sensitive
and which can provide a reliable reference for the standard of fraxetin quality control.
