Optimization of Diphasic Dynamic Extraction Technology for Quercetin from Flos Sophorae Immaturus
Objective: To optimize diphasic dynamic extraction technology of quercetin from Flos Sophorae Immaturus. Method: With the content of quercetin as index
based on single factor tests
L9(34) orthogonal test was adopted to optimize diphasic dynamic extraction technology of quercetin by taking extraction time
extraction temperature
extraction times and sulfuric acid concentration as factors.The content of quercetin was determined by HPLC
chromatographic conditions were as follows:Agilent ZORBAX C18 column(4.6 mm×250 mm
5 μm)
mobile phase of methanol-0.02% H3PO4(45:55)
detection wavelength 254 nm
injection volume 10 μL. Result: Optimal extraction technology was as following:extracted 2 times with 2% sulfuric acid at 85℃
2.5 hours per time;Yield of quercetin was up to 5.22 mg· g-1
which was more than ethanol extraction method of 3.26 mg· g-1. Conclusion: This optimized extraction technology was feasible and stable with high yield of quercetin.
