Investigation of Factors Influencing on Extraction Efficiency of Active Constituents in Ligustri Lucidi Fructus and Optimization of Ethanol Extraction Process of Zhenzhi Granules
Objective: To optimize ethanol extraction technology of Ligustri Lucidi Fructus and Herba Hedyotis from Zhenzhi granules. Method: Contents of oleanolic acid
ursolic acid and specnuezhenide were determined by HPLC.Factors influencing transfer rate of specnuezhenide were investigated by single factor tests
with contents of specnuezhenide and total triterpenoid acids as indexes
orthogonal test was adopted to optimize ethanol extraction technology of Zhenzhi granules by taking the amount and concentration of ethanol
extraction time and times as factors. Result: Optimum extraction process was as following:coarse crushed Ligustri Lucidi Fructus and mixed with Herba Hedyotis
extracting twice with 70% ethanol (9 folds for the first time
7 folds for the second time) for 1.5 h each time;Extraction amount of specnuezhenide and total triterpenoid acids were 19.083 and 7.163 mg·g-1
respectively. Conclusion: This optimized extraction process was reasonable
stable and feasible
which could provide scientific basis for industrial production of Zhenzhi granules.
