Determination of Oleanolic Acid and Hederagenin in Yanyan Granule by HPLC-ELSD
Objective: To establish a method for the determination of both oleanolic acid and hederagenin in Yanyan granule by high performance liquid chromatography and evaporative light scattering detection (HPLC-ELSD). Method: A HPLC-ELSD system was applied
using a Cosmosil PAQ-C18 (4.6 mm×250 mm
5 μm) column;mobile phase consisted of methanol (A)-water (B) under gradient elution;the flow rate was 0.9 mL·min-1. Result: As for oleanolic acid and hederagenin
the linear range was between 1.321-5.285 μg(r=0.999 3)and 0.473 6-1.894 μg(r=0.999 2)
with average recovery rate to be 98.5% and 100.2%
respectively. Conclusion: The method developed is simple
rapid with reliable results
which could be applied for the determination of oleanolic acid and hederagenin in Yanyan granule.
