Optimization of Purification Technology for Total Flavonoids from Saussureae Involucratae Herba by Macroporous Resin
Objective: To optimize purification technology of total flavonoids from Saussureae Involucratae Herba by macroporous resin. Method: Taking rutin as index ingredient
macroporous resin model was screened by static adsorption-elution test
single factor tests were adopted to investigate effects of sample amount
sample flow rate
elution rate and other factors on purification technology of total flavonoids from Saussureae Involucratae Herba. Result: X-5 macroporous resin showed the best separation efficiency by compared with other resins
its optimum purification technology were as follows:the concentration of sample solution 50 g·L-1
sample volume of 6 BV
adsorption rate of 3 BV·h-1
washed impurities with 3 BV of water
removed other purities with 6 BV of 20% ethanol
eluted with 5 BV of 40% ethanol
elution velocity of 3 BV·h-1
collected 40% ethanol eluent; After purified
the content of rutin increased from 1.07% in extract to 12.14% in ethanol eluent. Conclusion: X-5 macroporous resin could purify total flavonoids from Saussureae Involucratae Herba.
