Optimization of extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis by Response Surface Method
Objective:To optimize the extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.Methods:The extraction conditions of ratio of liquid to solid
extraction time and times were optimized by response surface method with the total alkaloids as the selective marker.The purification condition of total alkaloids was also studied.Results: The optimum extraction conditions were as follows: extraction repeated 4times
1 hour every time and ratio of liquid to solid was 18.125:1.The yield of total alkaloids under this condition was 66.44 mg·g
-1
The total alkaloids were concentrated by acetic ether extraction under pH 12
with the ratio 1:1 of solvent to extract
also the extraction was repeated 6 times
then the content of total alkaloids was 66.130%.Conclusion:The experiment provides a basis for improvement of extraction condition of total alkaloids from Rhizoma Coptidis and Cortex Phellodendri Chinensis.
