Extract of Ligustri Lucidi Fructus Inhibits Expression of Angiogenic Factors in Human Hepatocellular Carcinoma Cells
摘要
目的:研究女贞子提取物中熊果酸对人肝癌细胞生长抑制作用及对血管内皮生长因子(VEGF)
转化生长因子-α(TGF-α)表达的抑制作用
探讨其抗肿瘤机制。方法:女贞子提取物与PLC/PRF/5细胞孵育
以四甲基偶氮唑蓝法(MTT)检测其对人肝癌细胞PLC/PRF/5生长的抑制作用
并通过酶联免疫吸附实验(ELISA)测定细胞培养上清液中VEGF
TGF-α的表达;小鼠右前肢腋皮下接种瘤细胞
连续给药14 d后取瘤组织
检测小鼠肿瘤的抑制率
并以免疫组织化学方法检测瘤组织VEGF
TGF-α表达。结果:女贞子提取物中的熊果酸在体内和体外试验结果均显示抑制人肝癌细胞生长
并对VEGF
TGF-α表达有明显的抑制作用。结论:女贞子提取物具有抗肿瘤作用
抑制VEGF
TGF-α表达可能是其抗肿瘤作用机制之一。
Abstract
Objective:To study effects of Ligustri Lucidi Fructus on hepatocellular carcinoma cells(HCC) and its influence on vascular endothelial growth factor(VEGF)
transforming growth factor-alpha(TGF-α) and to discuss its mechanism of action.Method:We examined the inhibition of PLC /PRF /5 cells after incubation with the extract of L.lucidi Fructus by 3-[4
5-dimethylthiazol-2-yl]-2
5-diphenyltetrazolium bromide(MTT).We examined the expressions of VEGF
TGF-α by enzyme-linked immunosorbent assay(ELISA).PLC /PRF /5 tumor-bearing mice mode was established by the inoculation of cancer cell suspension subcutaneously in mice’s right anterior limbs.We examined inhibition rate of tumor in mice treated with L.lucidi Fructus for 14 d intragastrically.We further examined the expressions of angiogenic factors in HCC xenografts by immunohistochemical methods.Result:The extract of L.lucidi Fructus can inhibit the proliferation of tumor cell growth as well as the expressions of VEGF and TGF-α.Conclusion:The extract of L.lucidi Fructus has anti-tumor effect
and the release of VEGF and TGF-α might be one of it’s anti-tumor mechanism.
