Separation and Purification of Quercetin-3-O-glucoside from Lophatherum gracile by High-speed Counter-current Chromatography Coupled with UNIFAC Mathematical Model
Objective:To develop an economic and sensitive method for separation of quercetin-3-O-gluco-side from Lophatherum gracile.Method:High-speed counter-current chromatography(HSCCC) and UNIFAC math-ematic model were successfully used to separate the target component.The solvent system composed of ethyl acetate-n-butanol-water(2:1:3) was singly prepared by UNIFAC mathematic model
and the comparison of the results pro-duced by HSCCC of using the solvent system prepared by UNIFAC and the conventional method were carried out.Result:The quercetin-3-O-glucoside with purity > 99% was successfully separated from L.gracile by HSCCC using the singled prepared solvent system
which had no difference compared with the classic method for preparation of the solvent system
but much of ethyl acetate
n-butanol and water were saved.Conclusion:In the HSCCC separation of quercetin-3-O-glucoside from L.gracile
using the single prepared solvent system by UNIFAC mathematic model can not only save resource
