摘要

目的:建立轮枝拟青霉菌Paecilomyces verticillatus发酵菌丝体中虫草菌素含量测定的反相高效液相色谱方法。方法:样品用20%乙醇超声提取

采用Spheriorb C18柱（4.6 mm×150 mm

5μm）

以0.02 mol·L-1乙酸铵-甲醇（85∶15）为流动相

柱温30℃

流量1 mL·min

检测波长260 nm。结果:在上述色谱条件下

虫草菌素回收率为94.0%～98.3%

RSD为0.98%

最低检出浓度为0.01 mg·kg-1。测得菌丝体中虫草菌素含量为3.12 mg·kg-1。结论:方法简便、快速、分离效果好、准确可靠

可作为发酵生产中虫草菌素的质量控制监测手段。

Abstract

Objective:To develop a RP-HPLC method for determination of cordycepin in Mycaelia of Paecilomyces verticillatus.Method:The sample was extracted by 20% alcohol

detected by Spheriorb C18 column（4.6 mm×150 mm

5 μm）under 30 ℃.The mobile phase was methyl alcohol and acetic acid ammonium（0.02 mol·L-1） under the rational of 15:85.the flow rate was 1.0 mL·min-1

detection wavelengths was 260 nm.Result:The recovery rate of cordycepin was 94.0%-98.3% with RSD of 0.98%

the limit of detection was 0.01 mg·kg-1.The content of cordycepin in my caelia was 3.12 mg·g-1.Conclusion:It was proved that the RP-HPLC method was simple

rapid reliable

well-separated and precise

which could be used for quality control in formenting cordycepin.
