Effect of the Early Intervention with Didang Tang on Macrophages Regulation in Experimental Type 2 Diabetic Rats
Objective:To observe the effect of the early intervention with Didang Tang on the expressionlevels of macrophage colony-stimulating factor(M-CSF)
macrophage inflammatory protein-1a(Mip-1a) and matrix metalloproteinases-9(MMP-9) associated with macrophages in the experimental type 2 diabetic rats with the macrovascular lesions.Method: The experimental diabetic in rats was induced by using high fat diet and streptozotocin
then early
middle and late intenventions of Didang Tang(3.24 g · kg-1
ig) were applied to the rats.The mRNA expression of M-CSF and Mip-1a was detected by using Real-time PCR method
and the protein expression of MMP-9 was measured by using Western blot method in the aorta.Result: Compared with model group
the mRNA expression of M-CSF in the aorta were lower in the early Didang Tang group and rosiglitazone group(P<0.05).Compared with model group
the mRNA expression of Mip-1a in the aorta was lower in the early Didang Tang group
rosiglitazone group and simvastatin group(P<0.05).Compared with model group
the protein expression of MMP-9 in the aorta was lower in the early Didang Tang group
the middle Didang Tang group and rosiglitagone group(P<0.05).Conclusion: The mRNA expression of M-CSF and Mip-1a and the protein expression of MMP-9 in the aorta of the experimental type 2 diabetic rats reduced after early intervened with Didang Tang.The early intervention with Didang Tang could regulate the role of macrophages in the formation and stability of atherosclerotic plaque
and delay the development of diabetic macroangiopathy.
