摘要

目的：研究柴胡疏肝散醇提物的体外自由基清除能力和体内抗氧化活性。方法：体外抗氧化实验测定了柴胡疏肝散醇提物对1，1-二苯基-2-苦肼基自由基（DPPH）、羟自由基（·OH）和超氧阴离子自由基（O2-·）清除能力；体内抗氧化实验，小鼠随机分为正常组、模型组、柴胡疏肝散水提组（8.2 g·kg-1）、柴a组（1.6 g·kg-1）、柴胡疏肝散醇提物低、中、高剂量组（4.1，8.2，20.5 g·kg-1），除正常组外，其余组小鼠每天接受不可预见性温和应激，共28 d。造模1周后灌胃给药，研究柴胡疏肝散醇提物对小鼠的肝组织中过氧化氢酶（CAT）、超氧化物歧化酶（SOD）活性的影响，并与水提物做比较，评价其体内抗氧化活性。结果：体外抗氧化实验表明，柴胡疏肝散醇提物对DPPH，·OH和O2-·的清除率随着浓度的增大而增大，其对自由基清除率达到半数时的抗氧化剂浓度（EC50）分别为0.75 mg·L-1，35.01 g·L-1，52.17 g·L-1；体内实验表明，模型组小鼠肝脏中SOD，CAT酶活性显著低于正常组（P<0.01）。给药剂量越大，SOD，CAT酶活性越高，在本实验设定的给药浓度范围内，呈现一定的量效关系。与水提组相比，具有同等生药量的醇提物8.2 g·kg-1组SOD，CAT酶活性显著较高（P<0.05，P<0.01），说明醇提物的抗氧化作用较水提物高。结论：柴胡疏肝散醇提物具有较强的抗氧化生物活性。

Abstract

Objective: To investigate in vitro radical scavenging activity and in vivo antioxidant activities of ethanol extracts from Chaihu Shugan powder (CSPEE). Method: The antioxidant activities of CSPEE in vitro were measured by the methods of 1

and superoxide anion radicals scavenging capacity. The antioxidant activities of CSPEE was studied in vivo

which is compared with the aqueous extracts of Chaihu Shugan powder. Kunming mice were used to study the catalase (CAT) and superoxide dismutase(SOD) activities. Result: The antioxidant activities in vitro showed that free radical scavenging activity of CSPEE was higher. In the results of the animal experiments

the CAT and SOD activities in liver tissue were increased by CSPEE. It was suggested that the antioxidant ability of CSPEE was better than the aqueous extracts at the same drug concentration. Conclusion: The CSPEE owns good antioxidant activities.
