Effect of Maimendong Decoction on Apoptosis, Cell Cycle and Expression of EGFR and STAT3 Genes of a Non-small Cell Lung Cancer A549 Cell Line
目的:通过体外实验观察麦门冬汤对非小细胞肺癌A549细胞凋亡、细胞周期以及表皮生长因子受体(epidermal growth factor receptor,EGFR)、信号转导子和转录激活子3(signal transducer and activator of transcription 3,STAT3)基因表达的影响,探索中药复方麦门冬汤抑制非小细胞肺癌的作用靶点。方法:选取对数生长期A549细胞(2×104个/mL),倒置显微镜下观察麦门冬汤(0.25,2.5,25 g·L-1)作用24,48,72 h后A549细胞的形态学变化;4,6-二脒基吲哚(DAPI)荧光染色观察麦门冬汤(2.5,25 g·L-1)作用48 h后A549细胞的凋亡;流式细胞术结合PI染色法分析麦门冬汤(2.5 g·L-1)作用48 h后A549细胞的周期;实时荧光定量RT-PCR技术对A549凋亡细胞EGFR,STAT3基因表达进行半定量。结果:0.25,2.5,25 g·L-1麦门冬汤均可抑制A549细胞生长,使其发生不同程度的形态学改变;0.25,2.5 g·L-1麦门冬汤作用A549细胞48 h后,荧光显微镜下可见细胞核变形,染色质聚集、边缘化等凋亡形态学特征,流式检测显示细胞被抑制于G0/G1,G2/M期,RT-PCR结果提示细胞中EGFR,STAT3基因的表达量明显降低(P<0.01)。结论:麦门冬汤可抑制非小细胞肺癌A549细胞生长,诱导其凋亡,并将其阻滞于G0/G1,G2/M期,这可能与麦门冬汤下调EGFR,STAT3基因表达有关。

Objective:To observe the effect of Maimendong decoction on apoptosis
cell cycle and expression of epidermal growth factor receptor(EGFR) and signal transducer and activator of transcription 3 (STAT3) genes of lung a cancer cell A549 by experiments in vitro.
