摘要

目的：提取分离、纯化具有免疫抑制活性的黄芪糖蛋白（AmGP-2）并考察其理化性质、组成成分。方法：黄芪经水浸提，盐析分离，sevage试剂脱游离蛋白等方法提取糖蛋白，采用阴离子色谱柱和凝胶色谱柱进行纯化。通过苯酚-硫酸法、BCA、Molish试验、茚三酮反应、气相色谱分析、氨基酸分析、SDS-PAGE、UV、IR、β-消去反应等分析黄芪糖蛋白的理化性质及组成成分。结果： AmGP-2易溶于水，等电点约4.7，蛋白质、糖类成分质量分数依次为56.7%，29.3%，相对分子质量约31 kDa；含有18种氨基酸，其中天冬氨酸、谷氨酸、赖氨酸含量较高；糖链部分含有阿拉伯糖、葡萄糖及2种未确定的单糖。结论： AmGP-2具有显著的免疫抑制活性，为中药免疫抑制剂的开发提供参考。

Abstract

Objective: To extract and purity Astragali Radix glycoprotein(AmGP-2) having immunosuppressive activities

then investigate its physicochemical properties and composition. Method: AmGP-2 was handled by water extraction

sevage method;Then purified by anion and gel chromatography.Physicochemical properties and composition of AmGP-2 were analyzed by phenol-sulfuric acid method

IR and β-elimination reaction. Result: AmGP-2 was soluble in water with isoelectric point(pI) of 4.7 and relative molecular weight about 31 kDa

respectively;It contained 18 kinds of amino acids

higher content including aspartic acid

glutamic acid and lysine;Its carbohydrates chain portion contained arabinose

glucose and two kinds of undetermined monosaccharides. Conclusion: AmGP-2 had significant immunosuppressive activity

this study could provide a reference for development of immunodepressants of Chinese herbal medicines.
