摘要

目的：优选秦岭龙胆中齐墩果酸的乙醇回流提取工艺。方法：采用HPLC测定齐墩果酸含量，流动相乙腈-甲醇-水-磷酸-三乙胺（25：15：10：0.01：0.02），检测波长210 nm。在单因素试验基础上，根据Box-Benhnken中心组合设计原理，利用三因素三水平响应面分析法考察料液比、提取次数、提取时间对齐墩果酸提取量的影响。结果：最佳提取工艺为料液比1：12，提取数2次，提取时间76 min；齐墩果酸提取量0.583 mg·g-1，与预测值0.605 mg·g-1的偏差仅3.64%。结论：采用响应面法优选的秦岭龙胆回流提取工艺稳定可行，为秦岭龙胆的资源开发及临床推广提供参考。

Abstract

Objective: To optimize ethanol reflux extraction process of oleanolic acid in Gentiana apiata. Method: HPLC was employed to determine the content of oleanolic acid with mobile phase of acetonitrile-methanol-water-phosphoric acid-triethylamine(25:15:10:0.01:0.02) and detection wavelength at 210 nm.On the basis of single factor tests
