Optimization of Water Extraction and Alcohol Precipitation Technology for Hemorrhoid Fumigants by Orthogonal Test
Objective: To optimize water extraction and alcohol precipitation technology of hemorrhoid fumigants. Method: With composite score of the content of total alkaloids and dry extract yield as index
effects of the amount of water
extraction times and time on extraction technology was investigated by orthogonal test;with the content of total alkaloids as index
orthogonal test was adopted to investigate effects of relative density of extract
concentration and time of alcohol precipitation on alcohol precipitation technology.UV was employed to determine the content of total alkaloids with detection wavelength at 416 nm. Result: The best water extraction and alcohol precipitation technology was as following:decocted twice with 10 times the amount of water for 1.5 h per time
relative density of extract of 1.06 g·mL-1 at 25 ℃
alcohol precipitation concentration 60%
storage time of 12 h;the concentration of total alkaloids was 2.040 g·L-1. Conclusion: These optimized technologies were stable and feasible to provide a reference for standardized production of hemorrhoid fumigants.
