Determination of Three Glycosides and Gastrodine Contents in Gastrodiae Rhizoma by RP-HPLC
Objective: The study was performed to establish the RP-HPLC gradient elution method for determination of three glycosides and gastrodine contents in Gastrodiae Rhizoma. Method: RP-HPLC gradient elution method was used. The extracts were separated on a ZORBAX-SB-C18 analytical column(4.60 mm×250 mm
5 μm) with CH3OH-0.1% phosphoric acid as mobile phase
the UV detection wavelength was set at 270 nm
the flow rate fixed at 0.6 mL·min-1and the column temperature was maintained at 25 ℃. Result: The linear ranges of gastrodine
uridine
guanosine and adenosine were 0.286-4.29 μg(r=0.999 9)
0.028 9-0.433 5 μg(r=0.999 7)
0.066 8-1.002 μg(r=1.000 0)
0.135 6-2.034 μg(r=1.000 0)
respectively. Conclusion: This method is fast
simple and accurate
which can be used for quality control of Gastrodiae Rhizoma.
