Influence of Endothelial Function and Blood Rheology in Diabetic Rats by Paeonol and Tanshinol
Objective: To explore the influence of endothelial function and blood rheology in diabetic rats by paeonol and tanshinol. Method: Diabetic model was established by streptozotocin
the modeling 40 rats were randomly divided into model group
paeonol group(2.5 mg·kg-1
ig)
tanshinol group(5 mg·kg-1
ig)
paeonol combined with tanshinol group. Another 10 healthy rats were taken as normal control group
normal group and model group mice were given distilled water
the administration lasted 8 weeks. Body weight
blood sugar
the endothelial function and hemorheology index changes of rats were determined. Result: ①Compared with model group
there was no significant difference for the blood sugar in the phenol group
tanshinol group and combined group
compared with normal group
there was no significant difference. ②Compared with normal group
nitric oxide(NO)
endothelin(ET-1)
von Willebrand's factor(vWF) and plasminogen astivator inhibitor-1(PAI-1) were increased in model group (P<0.05)
after 8 weeks treatment
compared with model group
there were no significant difference for vWF and NO in the phenol group
tanshinol group
NO in combined group significantly increased (P<0.05)
vWF decreased (P<0.05);there were no significant for ET-1 in the phenol group
and ET-1 of tanshinol group and combined group significantly increased (P<0.05). There were significant difference for PAI-1 in the phenol group
tanshinol group and combined group(P<0.05);fibrinogen(Fg) in the phenol group had no significant differences
and Fg in tanshinol group and combined group had significant difference (P<0.05). ③Compared with normal group
in model group
whole blood low shear viscosity
whole blood high shear viscosity
plasma viscosity
erythrocyte and fibrin deposition pressure had significant difference (P<0.05). Compared with model group
the whole blood viscosity and high and low shear in the phenol group
tanshinol group and combined group were significantly lower (P<0.05)
red blood cells were significantly lower (P<0.05)
red blood cells of deposited in joint group were higher than that of group (P<0.05). Fibrous protein in tanshinol group and combined group was significantly lower than that of model group (P<0.05). Conclusion: Paeonol combined with tanshinol can improve the blood rheology of experimental diabetic rats
has certain protective effect on endothelial function of diabetic rats.
