Optimization of Extraction Technology for Total Saponins from Ziziphi Spinosae Semen by Orthogonal Test
Objective: To optimize extraction technology of total saponins from Ziziphi Spinosae Semen and provide a reference for industrial production of this component. Method: HPLC-UV method was employed to detect contents of jujuboside A and jujuboside B with mobile phase of acetonitrile-water(34: 66) and detection wavelength at 204 nm.On the basis of single factor tests

extraction time and extraction times on extraction process were investigated by orthogonal test. Result: Linear ranges of jujuboside A and jujuboside B were 0.24-2.40

average recovery of them were 100.39% and 99.40%.The best extraction technology of total saponins from Ziziphi Spinosae Semen was as following:extracted 2 times with 12 times the amount of 70% ethanol for 1.5 h per time;average contents of jujuboside A and jujuboside B were 0.606

respectively. Conclusion: This established HPLC method is accurate and reliable to detect contents of jujuboside A and jujuboside B.At the same time
