Effect of Replenishing Kidney-essence and Removing Phlegm on Ability of Learning and Memory and Antioxidant in APP/PS1 Double Transgenic Mice
Objective: To investigate the effect of replenishing kidney-essence and removing phlegm on the ability of learning and memory and antioxidant in APP/PS1 double transgenic mice. Method: APP/PS1 double mice were randomly divided into model group
western medicine group(6.5×10-4g·kg-1·d-1)
Bushen Huatan Yizhi recipe (BSHTYZR) high dose group (15.6 g·kg-1·d-1)
low dose group (7.8 g·kg-1·d-1)
10 in each groups. Blank group mice (C57BL/6J) were 10 months of age with the same genetic background mice. After intragastrical administration for 3 months
the ability of learning and memory was assessed by the step down avoidance test and shuttle box test;colorimetric assay was used to measure mouse brain tissue superoxide dismutase (SOD) and malondialdehyde (MDA) content. Result: Step down avoidance test results showed that compared with model group
each group of mice significantly prolonged the incubation period and the number of errors decreased significantly (P<0.01);shuttle box test results showed that compared with model group
the number of active avoidance in treatment groups increased significantly (P<0.01
P<0.05)
and active avoidance incubation period
the passive avoidance period significantly shorten (P<0.01);shuttle box test results showed that compared with the model group
in the rest of the administration group the small active avoidance significantly increased the number of rats (P<0.01
P<0.05)
while the active avoidance latency and passive avoidance latency was shorter (P<0.01);in addition
in BSHTYZR high dose group and low dose group SOD activity of brain tissue increased significantly compared with the model group (P<0.01)
MDA was significantly lower than the model group (P<0.01). Conclusion: BSHTYZR has a certain improvement on APP/PS1 double transgenic mice learning and memory
