Objective: To optimize extraction process of four flavonoids in Scutellariae Radix and provide references for control extracts research of thin-layer chromatography of Scutellariae Radix. Method: Effects of ethanol concentration

baicalein and wogonin were investigated and forecasted by central composite design-response surface methodology.HPLC was adopted to determine contents of four flavonoids with mobile phase of acetonitrile-0.1% phosphoric acid solution and detection wavelength at 274 nm. Result: Optimal extraction process was as following:extracted twice with 12 times the amount of 54% ethanol for per time of 135 min.Yields of baicalin

respectively.Bias between the predicted and observed value of overall desirability was 1.19%. Conclusion: This optimized extraction technology is stable and feasible with good prediction.
