Objective: To optimize deproteinization process of total polysaccharides from exocarp of unripe fruits of Juglans mandshurica. Method: With retention rate of total polysaccharides and deproteinized rate as indexes

trichloroacetic acid(TCA)

enzyme-Sevage and enzyme-TCA on deproteinization of total polysaccharides were compared and optimized by single factor tests and orthogonal test. Result: Effect of papain-TCA method to take off protein was the best

the concentration of TCA 6%.Retention rate of total polysaccharides and deproteinized rate were 82.48% and 84.76%
respectively. Conclusion: Deproteinized effects on total polysaccharides from exocarp of unripe fruits of J. mandshurica have significant difference by different deproteinized methods
enzyme-TCA combination method is easy to operate with optimum deproteinized effect and short time-consuming.
