Effect of Tiaoganshen Qutanyu-formula on Transforming Growth Factor-, Connective Tissue Growth Factor and Fibronectin in Spontaneous Hypertensive Rats
Objective: To observe the effect of Tiaoganshen Qutanyu-formula (TQF) on transforming growth factor-β1 (TGF-β1)
connective tissue growth factor (CTGF) and fibronectin (FN) in kidney tissue in spontaneous hypertensive rats (SHR) administered before blood pressure peak. Method: A total of 30 12-week male SHR were randomly divided into 3 groups:the TQF group (4 g · kg-1)
the losartan group (30 mg · kg-1) and the model group. Another healthy 10 SD rats were taken as the normal group. The mRNA and protein expressions of TGF-β1
CTGF and FN were detected by using RT-PCR and Western Blot after 12 weeks of treatment. Result: Compared with the normal group
the mRNA expressions of TGF-β1
CTGF and FN were obviously up-regulated
protein expressions of CTGF and FN were also up-regulated in the model group (P<0.01).Compared with the model group
mRNA and protein expressions of TGF-β1
CTGF and FN were down-regulated in TQF and losartan groups (P<0.05
P<0.01). Moreover
the results were lower in the TQF group (P<0.05). Conclusion: TQF taken before blood pressure peak could reduce the aggregation of extra celluar matrix and prevent kidney tissue injury in SHR. The mechanism might be the synergistic effect of Yin-yang theory of the expression inhibition of TGF-β1
