Abstract
Objective: To establish an ultra performance liquid chromatography (UPLC) method for the simultaneous determination of five lignans (schisandrin
schizandrol B
schisantherin A
deoxyschizandrin and schisandrin B) in Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus
and compare the contents of lignans in Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus from different origins. Method: The chromatographic separation was performed on Waters Acquity UPLC BEH C18 (2.1 mm×50 mm
1.7 μm) with the mobile phase of acetonitrile and water at a flow rate of 0.4 mL·min-1. The detection wavelength was 235 nm
the column temperature was 45 ℃
and the injection volume was 1 μL. With the contents of the five lignans as the evaluation index
the clustering analysis was used to identify the species of Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus. Result: Within the analytical time of 10 minutes
the five lignans showeda good separation and linear relationship at a range of 0.001-0.222 μg
with correlation coefficients of more than 0.999 8.Their average recoveries ranged between 97.7% and 105.1% (RSD<3.0%). The contents of five lignans were significantly different in Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus. Moreover
the cluster analysis results also indicated that they could be classified into two groups. Conclusion: The method is highly reproducible
accurate and sensitive and so can provide reference for the classification and quality control of Schisandrae Sphenantherae Fructus and Schisandrae Chinensis Fructus.
