Determination of Procyanidin B-2 and Procyanidin C-1 in with UPLC
Objective: To established an UPLC method for simultaneously determination of two indicative components of Cinnamomum (Lauraceae)
namely procyanidin B-2 and procyanidin C-1
and to complete the determination of 17 batches of cinnamon bark samples by the established method. Method: The UPLC analysis was performed on an shiseido capcell pak C18 column (2.0 mm×100 mm
2μm) with acetonitrile(A)-0.2% acetic acid solution(B) as mobile phase for gradient elution(0-8 min
11%-20%A;8-10 min
20%-25%A;10-12 min
25%-75% A;12-13 min
75%A). The detection wavelength was set at 280 nm. The flow rate was 0.35 mL·min-1. The column temperature was set at 25℃. Result: Two indicative components showed a wide linear range and a good linear relationship. The content of the two components by UPLC is moderately correlated with the content of total polyphenol by Ultraviolet-visible Spectrophotometry in the 17 batches of cinnamon bark samples from market. Conclusion: The established method is simple
accurate and reproducible
and can be applied to determine the content of water-soluble oligomers procyanidin in cinnamon bark.
