目的:建立川芎-天麻药对中天麻素、天麻苷元、阿魏酸和藁本内酯4种成分的含量测定方法。方法:采用Inertsil ODS-3色谱柱(4.6 mm×250 mm
5 μm)
流动相甲醇-0.05%磷酸水梯度洗脱
检测波长220 nm(天麻素、天麻苷元)
320 nm(阿魏酸、藁本内酯)
柱温25℃
流速1.0 mL·min-1。结果:4种效应成分分离度良好
标准曲线在检测范围内线性关系良好
天麻素、天麻苷元、阿魏酸和藁本内酯的平均回收率分别为104.0%
103.4%
100.6%
101.8%
RSD分别为0.4%
0.5%
4.5%
2.6%。结论:该方法简便、精密度、重复性好
可用于川芎-天麻药对中4种效应成分的含量测定。
Abstract
Objective: To establish a method for the content determination of gastrodin
4-hydroxybenzyi aicohol
ferulic acid and ligustilide in extracts of Ligusticumc chuanxiong-Gastrodia elata. Method: The determination was performed on Inertsil ODS-3 chromatographic column (4.6 mm×250 mm
5 μm) with mobile phase of methanol-0.05% phosphoric acid for gradient elute
at the flow rate of 1.0 mL·min-1. The detection wavelength was set at 220 nm (gastrodin
4-hydroxybenzyi aicohol)
320 nm (ferulic acid
ligustilide) and column temperature was 25℃. Result: The four compounds were well separated and showed good linearity within the concentration ranges tested. Their average recovery rate was 104.0%

100.6% and 101.8% for gastrodin
4-hydroxybenzyi aicohol
ferulic acid and ligustilide;and RSD of the above four components was 0.4%

4.5% and 2.6%
respectively. Conclusion: This method is simple
precise and repeatable
and it can be used to determine the four active components in Ligusticumc chuanxiong-Gastrodia elata.
