Identification of Constituents in Mori Folium Aqueous Extract and Drug Serum
Objective: To analyze and identify the chemical compounds in Mori Folium aqueous extract and the components in serum of rats after oral administration. Method: HPLC was employed to analyze the compounds in Mori Folium aqueous extract as well as in rat serum after intragastric administration. LTQ-Orbitrap XL high-resolution mass spectrometer was used for online ions detection and secondary ions scanning to identify the compounds in Mori Folium aqueous extract and its metabolites in blood samples. Result: The 11 chemical compounds were identified in the Mori Folium aqueous extract
including isochlorogenic acid(1)

7-dlhydroxycommarin-7-O-β-D-glucopyranoside(2)
scopolin(3)
chlorogenic acid(4)
kaempferol-3
7-dl-O-β-D-glucopyranoside(5)
4-caffeoylquinic acid methyl ester(6)
rutin(7)
hyperoside(8)
isoquercitrin(9)
astragalin(10)
and isorhamnetin-3-O-glucoside(11). The 30 compounds were preliminarily identified in the blood
and major of
them were aglycone metabolites. The above 11 original compounds were not found in serum. Conclusion: The present method can be used for the identification of the compounds in Mori Folium aqueous extract and blood of rats after oral administration
providing reference for further efficacy study and secondary development of Mori Folium.
