摘要

目的：探讨健脾温胃散对脾气虚胃炎大鼠血清肿瘤坏死因子-α（TNF-α），白细胞介素-6（IL-6）及胃组织白细胞介素-1受体相关激酶-4（IRAK-4）mRNA表达的影响，为中药方剂治疗脾气虚胃炎提供实验依据。方法：Wistar大鼠60只，随机分正常组，胃炎模型组，阳性药物组（丽珠得乐胶囊，0.016g·kg-1·d-1）及健脾温胃散高、中、低剂量组（0.324，0.259，0.194g·kg-1·d-1），除正常组外均建立大鼠脾气虚胃炎动物模型，测量各组大鼠的肛温变化，各组大鼠ig相应药物30d治疗后，采用放射免疫法测定其血清中IL-6，TNF-α的含量；苏木素-伊红（HE）染色观察胃组织病理学情况；逆转录-聚合酶链式反应（RT-PCR）方法观察健脾温胃散对胃炎大鼠模型胃黏膜IRAK-4mRNA表达的影响。结果：与正常组比较，模型组大鼠肛温变化显著降低，大鼠血清IL-6，TNF-α含量显著升高，大鼠胃组织IRAK-4mRNA表达显著升高（P<0.01），大鼠胃黏膜的病理损伤较明显；与模型组比较，健脾温胃散高、中、低剂量组能明显改善模型大鼠的证候特征及肛温变化，改善胃黏膜的病理形态，能显著降低胃炎模型大鼠血清IL-6，TNF-α含量（P<0.01），下调胃组织IRAK-4mRNA表达（P<0.01）。结论：健脾温胃散能使模型大鼠的肛温恢复正常，改善胃黏膜的病理形态，降低脾气虚胃炎模型大鼠血清IL-6，TNF-α含量，抑制IRAK-4表达，可能是其治疗胃炎的作用机制之一。

Abstract

Objective: To investigate the effect of Jianpi Wenwei powder on the expression of serum tumour necrosis factor-α(TNF-α)

interleukin-6(IL-6) and interleukin-1 receptor-associated kinase 4(IRAK-4) mRNA in the gastric mucosa of spleen-deficiency rats. Method: Totally 60 Wistar rats were randomly divided into normal control group

and inhibit the expression of IRAK-4

which may be one of its mechanisms for treatment of gastritis.
