摘要

目的：采用超高效液相色谱-电喷雾飞行时间串联质谱法（UHPLC-ESI-Q-TOF-MS/MS）对紫花地丁水提取物的主要化学成分进行分析鉴定。方法：采用Welch C18色谱柱（2.1 mm×100 mm，1.7 μm），以0.1%甲酸水（A）-乙腈（B）为流动相梯度洗脱，流速0.35 mL·min-1，质谱采用电喷雾（ESI）离子源，在正负离子模式下采集数据，运行时间为30 min。结果：通过二级高分辨质谱分析结合对照品数据及相关文献，共鉴定32个化合物，包含香豆素、黄酮及有机酸类成分。结论：该研究利用UHPLC的高效分离能力和MS的高灵敏度检测优势，建立的分析方法，为控制紫花地丁药材质量、临床疗效及阐释其作用机制提供了科学依据。

Abstract

Objective: Using the UHPLC-ESI-Q-TOF-MS/MS to analyze and identify the the main components in the water extract of Violae Herba. Method: The separation was performed on UHPLC Welch C18 column (2.1 mm×100 mm

1.7 μm)

with a mobile phase using water containing 0.1% formic acid (A) and acetonitrile (B) for gradient elution

the flow rate was 0.35 mL·min-1. Electrospray ion (ESI) source were applied for the analysis under the positive/negative ion mode

and the running time was 30 min. Result: Based on the standards of MS/MS and compared with the reference results

containing coumarins

flavonoids and organic acids were identified. Conclusion: The efficient separation ability of UHPLC and high sensitive detection of MS are used in this study

which will provide the evidences for evaluating the quality of Violae Herba

stabilizing the curative effect in clinic

and explaining the mechanism.
