Simultaneous Determination of 7 Components in Salviae Miltiorrhizae Radix et Rhizoma by UPLC-MS/MS
Objective: To establish a UPLC-MS/MS (Ultra performance liquid chromatography tandem mass spectrometry) method for simultaneous determination of 7 components in Salviae Miltiorrhizae Radix et Rhizoma:protocatechualdehyde
salvianolic acid B
rosmarinic acid
tanshinone Ⅰ
tanshinone ⅡA
cryptotanshinone and dihydrotanshinone. Method: The separation and analysis were performed on Hypersil Gold C18 (2.1 mm×100 mm
1.9 μm)
with 0.1% acetic acid solution-methanol as the mobile phase for gradient elution. The column temperature was set at 30℃ with flow rate of 0.3 mL·min-1. Electrospray ionization (ESI) source was applied and operated in alternating positive and negative mode. Selective reaction monitoring (SRM) mode was used to quantify the 7 compounds. Result: The 7 compounds showed good linear relationship in the range of 1-460 μg·L-1. The average recovery rates were 83.41%-116.05%; the detection limits were 0.046-0.5 μg·L-1 and the relative standard deviation was less than 4.7%. Conclusion: This method is rapid
accurate
reproducible and sensitive
and has been successfully applied in the simultaneous quantification of 7 components in Salviae Miltiorrhizae Radix et Rhizoma. It can be used for the study of Salviae Miltiorrhizae Radix et Rhizoma and its prescriptions as well as for quality control.
