Objective: To design and establish UPLC fingerprint on several varieties of Pueraria from Sichuan
and provide basis for its medicinal materials identification and quality control. Method: The UPLC fingerprint was performed on Agilent Zorbax SB-C18 (2.1 mm×5 mm
1.8 μm)
with acetonitrile -0.1% acetic acid aqueous solution as mobile phase; the column temperature was 30℃; the flow rate was 0.4 mL·min-1 and injection volume was 0.5 μL; the detection wavelength was set at 250 nm. In addition
similarity analysis and cluster analysis were conducted. Result: The UPLC fingerprint of P. lobata
P. thomsonii and P. omeiensis were established
and respectively confirmed 29
18 and 16 peaks. Seven peaks in P. lobata and six peaks in P. thomsonii and P. omeiensis were identified by comparing the retention time of reference substance and chromatographic peak. The main differences between three varieties were peaks 2

which were not detected in P. thomsonii and P. omeiensis; the main differences between P. thomsonii and P. omeiensis were peaks 6 and 14
which were not detected in P. omeiensis. The correlation results obtained in clustering analysis were close to similarity analysis results. Conclusion: UPLC fingerprint technology is fast and its result is reliable and accurate
so it can be used as a reliable method for study on this kind of medicine fingerprint.
