Effect of Xuefu Zhuyu Capsule in Regulating Jagged1/Notch Signal in Angiogenesis
Objective: To explore the effect of Xuefu Zhuyu capsule (XZC) in regulating Jagged1/Notch signal in angiogenesis. Method: XZC-containing serum and blank serum were used respectively to treat human microvascular endothelial cell-1 (HMEC-1). The mRNA expression of Jagged1 was detected by Real-time quantitative (PCR) at 12

and the protein expression of Jagged1 was detected by Western blot at 12

48 h. The γ-secretase inhibitor DAPT was used to block the Notch signaling pathway under the optimal condition of promoting angiogesis by XZC. The cells were randomly divided into 3 groups
namely DAPT processing group
solvent control group and blank control group. The angiogenesis effect was tested by an in vitro angiogenesis assay. Result: XZC up-regulated the mRNA expression of Jagged1 after culturing HMEC-1 cells for 12 h (P<0.01)
and the mRNA and protein expressions of Jagged1 were up-regulated at 24 h (P<0.05). The ability of angiogenesis in vitro was inhibited after the Notch signaling pathway was blocked by DAPT. Conclusion: XZC can regulate the expression of Jagged1
and Jagged1/Notch signaling pathway may be correlated with XZC's effect on regulating angiogenesis.
