Analysis on L-F and K Genetic Relationship of var. in Guizhou Province
目的:探讨贵州省宽叶缬草在叶绿体基因trnL-F,matK上的遗传关系。方法:利用trnL-F,matK序列对宽叶缬草进行序列分析,运用Bioedit,MEGA 5.0软件计算贵州省宽叶缬草居群遗传距离,并进行聚类分析建立系统发育树。结果:不同居群宽叶缬草trnL-F序列长度为947~985 bp,鸟嘌呤和胞嘧啶质量分数为35.92%~36.55%,其中差异位点8个,变异位点5个,居群间的遗传距离为0.000 0~0.005 4,最小进化法(minimum-evolution,ME),最大似然法(maximum likelihood,ML),邻接法(neighbor-joining,NJ),非加权配对算数平均法(unweighted pair group method with arithmetic mean,UPGMA)4种聚类构树结果显示15(江口县官和乡泗渡村寨上组),18(江口县凯德镇育苗基地)号样品明显与大部分的宽叶缬草样品区分出来;不同居群宽叶缬草matK序列长度为1 080~1 088 bp,鸟嘌呤和胞嘧啶质量分数为35.19%~36.64%,差异位点2个,变异位点1个,简约信息位点1个,居群间的遗传距离为0.000 0~0.000 9,ML,NJ,ME,UPGMA 4种聚类构树结果显示2(剑河县太拥乡太平村),5(剑河县岑松镇稿旁村),6(剑河县关么乡苗岭村),20(江口县闵孝镇渔凉溪村)号样品聚为一类,其余19份样品聚为一类。结论:贵州省宽叶缬草居群内trnL-F,matK序列具有高度保守性。
Abstract
Objective:To explore the genetic relationship of the Valeriana officinalis var. latifolia in Guizhou province at trnL-F and matK sequence. Method:The trnL-F and matK sequences of V. officinalis var. latifolia were analyzed. The genetic distance was calculated by Bioedit

and then the clustering analysis was conducted to establish the phylogenetic tree. Result:In the different populations of the V. officinalis var. latifolia
the lengths of trnL-F sequence ranged from 947-985 bp
and the guanine and cytosihe mass fraction was between 35.92%-36.55%
including 8 differential sites and 5 variable sites. The genetic distance ranged from 0.000 0-0.005 4.The branch type of phylogeny trees was constructed by ML
NJ
ME and UPGMA methods and showed that samples No.15 and 18 could be distinguished from the most samples. In the different populations of the V. officinalis var. latifolia
the lengths of matK sequence ranged from 1 080-1 088 bp
the guanine and cytosine mass fraction was between 35.19%-36.64%

6 and 20 were clustered as a class and the remained 19 samples were clustered into another group. Conclusion:The trnL-F and matK sequences of V. officinalis var. latifolia in Guizhou province are highly conservative.
