Effect of Wangbipian on Morphological Changes of Knee-joint of Animal with Osteoarthritis
目的:观察尪痹片对骨性关节炎模型动物膝关节组织病理损伤的影响。方法:家兔和大鼠模型动物均随机分为6组(假手术组,模型组,阳性药盐酸氨基葡萄糖组和尪痹片高、中、低剂量组)。除假手术组外,其余各组均造模,兔注射木瓜蛋白酶造模方式为尪痹片高、中、低剂量组(10.0,5.0,2.5 g ·kg-1)和盐酸氨基葡萄糖组(0.26 g ·kg-1),给药体积均选择为4 mL ·kg-1,造模40 d后连续灌胃给药33 d;大鼠切除前交叉韧带造模方式为尪痹片高、中、低剂量组(14.0,7.0,3.5 g ·kg-1)和盐酸氨基葡萄糖组(0.36 g ·kg-1),给药体积均为10 mL ·kg-1,造模60 d后连续灌胃给药31 d。每天给药1次,假手术组和模型组给予等体积水。给药结束后,观察动物膝关节活动度和膝关节周长,分别取膝关节进行病理检查,按照半定量分级标准进行综合评分和统计。结果:与模型组比较,尪痹片不同剂量组均可明显改善模型家兔膝关节的活动度,显著减少膝关节周长;可明显改善模型大鼠的活动状态;均可明显抑制2种模型动物膝关节滑膜细胞增生和炎性细胞浸润的程度。结论:尪痹片对木瓜蛋白酶注射诱导家兔和前交叉韧带切除诱导大鼠骨关节炎动物膝关节的软骨、滑膜损伤均具有明显改善作用,提示该制剂对骨关节炎具有一定的治疗作用。
Abstract
Objective:To explore the effect of Wangbipian on the histopathological lesion of knee-joint in experimental animal with osteoarthritis. Method:The therapeutical effect of Wangbipian on the osteoarthritis was detected in the rat osteoarthritis model induced by transection of anterior cruciate ligament on the knee-joint and the rabbit osteoarthritis model induced by injection of papain in knee-joint.SD rats and rabbits were divided into sham operation group

circumference and histopathological changes in the knee-joint were observed after the administration.The lesions of cartilago articularis
synovium and the inflammation of soft tissue were comprehensive evaluated. Result:As compared with the model group
administration of Wangbipian could significantly improve the ROM and decrease the circumference of the knee-joint.Histopathological analyses demonstrated that Wangbipian could inhibit the hyperplasisa of synoviocyte and reduce the infiltration of inflammatory cells in both rat and rabbit models. Conclusion:Wangbipian can alleviate the damage of cartilage and synovium of knee-joint in the experimental animal models of osteoarthritis
which suggests that Wangbipian has therapeutic effect on osteoarthritis.
